Effects of Culture Conditions on Dormant Status of In Vitro Regenerated Lilium Oriental Hybrid Bulblets

Abstract

|
This study was performed to investigate the effect of culture conditions on dormancy status of in vitro bulblets of Lilium Oriental hybrid ¢¥Casablanca¢¥ and ¢¥Marcopolo¢¥ . The regenerated bulblets were dormant regardless of culture period and sucrose concentration. Non-dormant bulblets were produced much larger at lower temperatures than 15¡É : 100¡É in ¢¥Casablanca¢¥, but were 50¡É and 70¡É at 10¡É and 15¡É, respectively in ¢¥Marcopolo¢¥. GA©ý was effective for production of non-dormant bulblet in vitro. This effect was greater in ¢¥Marcopolo¢¥ than ¢¥Casablanca¢¥. High concentration (9-12%) of sucrose in culture medium was effective for production of leafless bulblets and for bulblet growth. Dormancy of bulblets obtained in vitro was breaked completely by chilling at 4¡É for 6 weeks. But the chilling-treated bulblets were not bolted regardless of light and planting depth.
